Engine Knocking

So, i've been redoing my stereo system and haven't been driving the sc400 for a few weeks. I go to crank it up and it doesn't start. Try it again and the engine comes to life shaking, and knocking loudly. I shut it off and ignore for another day or so. I try it again and the same thing. I let it run a little longer the third time and the load knock goes away, but i think it has still has a slight knock in it, i haven't taken it on teh raod yet cause i'm scared. Is it a stuck valve or something, should i just drive it hoping it will smooth out? The car only has 97000 miles on it, i've run amsoil synthetic the last 10000miles and the timing belt was changed at 63000miles, what the hell is happening?

I would say let the engine run for a while and see if the internals sound better.
